Agarose is a purified linear galactoglycan hydrophilic colloid derived from agar or agar-containing algae. It is a linear polymer composed of β-d-galactosyl (1-4) linked to 3, 6-dehydrated-L-galactosyl. As a gel reagent, it is commonly used for routine nucleic acid analysis by gel electrophoresis or blotting (such as Northern or Southern) and for protein applications, such as radiative immunodiffusion (RID) assay. The minimum gelation time for 1% agarose is 30 minutes. Q2: How large of a band can the high-resolution agarose separate? A: The resolution of high-resolution agarose and 3:1 agarose is both approximately 2% (that is, for a fragment of 30bp, it can separate 30*0.02; for a 100bp fragment, it is 100*0.02.) Q: Can low-melting-point agarose be used for pulsed-field gel electrophoresis? A: No. The requirements for pulsed field gel electrophoresis are that the electroosmosis should be very low and the intensity should be very high.
