Klenow (3'→5'exo-) DNA polymerase is a proteolytic product of E. coli DNA polymerase I, retaining the DNA polymerase activity of DNA polymerase I but losing the 5'→3' exonuclease activity. This enzyme has further removed the 3'→5' exonuclease activity through mutation. It can be used for DNA labeling with random priming and DNA sequencing by the dideoxy method, among other applications.
