JC-1 is an ideal fluorescent probe widely used for detecting mitochondrial membrane potential (ΔΨm). JC-1 dye accumulates in mitochondria in a potential-dependent manner, allowing for the detection of mitochondrial membrane potential in cells, tissues, or purified mitochondria. In healthy mitochondria, JC-1 aggregates in the mitochondrial matrix to form polymers that emit intense red fluorescence (Ex=585 nm, Em=590 nm); in unhealthy mitochondria, due to the decrease or loss of membrane potential, JC-1 exists only as a monomer in the cytoplasm, producing green fluorescence (Ex=514 nm, Em=529 nm). JC-1 is not only suitable for qualitative detection, as the color change can directly reflect the change in mitochondrial membrane potential, but also for quantitative detection, as the degree of mitochondrial depolarization can be measured by the ratio of red to green fluorescence intensity. For observation, simply use the conventional settings for observing red and green fluorescence. The commonly used concentration range of JC-1 for detecting mitochondrial membrane potential in cells is 1–20 µg/mL, with 10 µg/mL being suitable for many cell types.
