bFGF is a member of the fibroblast growth factor (FGF) family and is a growth factor and signaling protein encoded by the FGF2 gene. Members of the FGF family exhibit broad mitogenic and cell-survival activities and are involved in various biological processes, including embryonic development, cell proliferation, morphogenesis, tissue repair, tumor growth, and invasion. In normal tissues, bFGF is present in the basement membrane and in the extracellular matrix beneath vascular endothelial cells. bFGF (FGF-2) is an essential component of all embryonic stem cell and induced pluripotent stem cell (iPSC) culture media, where it plays a critical role in maintaining pluripotency. However, wild-type FGF-2 is prone to proteolytic degradation and aggregation, making it inherently unstable. As a result, it has a short half-life in culture media and requires frequent medium changes. Thermostable FGF-2 variants are engineered through specific amino acid substitutions that enhance stability without compromising biological activity.
