THP-1 cells are a human monocytic leukemia cell line and a classic in vitro model for studying macrophage function. In experiments, cytokine stimulation is a common method for inducing THP-1 cell differentiation. Cytokines such as IL-4 and IL-13 can promote the differentiation of THP-1 cells into macrophages and initiate specific signaling pathways, thereby regulating cell function. This method can simulate in vivo immunomodulatory processes and provide a convenient way to study the functions of macrophages in inflammatory responses, immunomodulation, and anti-tumor activities. Macrophages are classified into two main types: M1-type macrophages and M2-type macrophages. THP-1 cells are usually induced to differentiate into macrophages using phorbol ester (PMA), and then, in the continued presence of PMA: lipopolysaccharide (LPS) and IFN-γ are added and cultured for 48 hours to induce M1 polarization; IL-4 and IL-13 are added and cultured for 48 hours to induce M2 polarization. Finally, the cells are resuspended in serum-free RPMI-1640 basal medium without stimulants and PMA, and can be maintained for 72 hours. This kit is used to differentiate THP-1 cells into macrophages (M1 or M2 type). This kit contains five reagents: recombinant human IL-13, recombinant human IL-4, recombinant human IFN-γ, phorbol ester (PMA), and lipopolysaccharide (LPS). PMA is used to initially differentiate THP-1 monocytes into unpolarized M0 macrophages. IFN-γ and LPS initiate the core signals for M1 polarization.
