These three cytokines are essential key growth f actors for the culture of Naïve T cells. This combination minimizes the generation of terminally differentiated effector T cells and exhausted T cells, and the expanded cells possess stronger self-renewal capabilities and in vivo persistence. IL-2 is a potent T cell growth factor that drives clonal expansion. On the basis of IL-7/IL-15, it provides additional proliferation signals. IL-7 is a survival factor, and its main function is to maintain T cell basal metabolism and survival. It is crucial for Naïve T cells and T memory stem cells. It can drive homeostatic proliferation and help maintain the Naïve/T memory stem cell phenotypes such as CD45RA, CCR7, and CD62L.
