Fluid Thioglycollate Medium Fluid Thioglycollate Medium is used for determining the presence of aerobic and anaerobic bacteria in normally sterile materials. Pancreatic Digest of C asein is the nitrogenous source. Yeast Extract supplies B-complex vitamins. Dextrose is the fermentable carbohydrate. L-Cystine and Sodium Thioglycollate allow Clostridium spp. to grow in this medium even under aerobic conditions. The latter also neutralizes the toxic effect of mercurial preservatives and peroxides. Resazurin indicates increases in the oxygen content by shifting to pink-red. The small amount of Agar maintains a low redox potential to stabilize the medium. There is a choice of 3 sizes - 500g, 2kg, and 10kg. PREPARATION: Mix 29.8g of medium in one Liter of purified water until evenly dispersed. Heat with repeated stirring until boiling to dissolve completely. Distribute and autoclave at 121°C for 15 minutes. After storage of the medium, if the medium demonstrates oxidation (color change of indicator), reheat the medium once in boiling water or steam bath.
