How to Sample Correctly Identify the spot(s). Avoid obvious plant roots, rocks, manure, or recently amended areas. Remove surface debris. Brush away litter, mulch, and gravel. Sample to 0–15 cm (0–6 in) depth. Use a clean trowel, probe, or auger. Prepare composite sample (recommended). Combine up to 5 cores in the mixing bag, knead to homogenize. Preserve sample. Using the paper funnel, add soil into the vial to the marked fill line. Shake well. Mix soil with the DESS preservative to fully saturate. Pack & ship. Place the vial in the return box, seal, and drop at the appropriate shipping location. After We Receive Your Sample DNA extraction is performed immediately—already included in the kit price. We archive both the original tube and purified DNA for at least 3 months (longer on request, space permitting).
