Background Adiponectin, also referred to as adipocyte complement-related protein of 30 kDa (Acrp30), is a protein secreted predominantly by adipocytes and exerts broad paracrine and endocrine effects on metabolic homeostasis and inflammation. It plays a critical role in promoting adipocyte differentiation, enhancing fatty acid oxidation, and improving insulin sensitivity. Circulating adiponectin levels are inversely associated with obesity, type 2 diabetes mellitus, and atherogenesis, highlighting its potential protective role in metabolic and cardiovascular disorders. Structurally, adiponectin oligomerizes into ~70 kDa noncovalently linked trimers, which are stabilized through disulfide bonds to form medium-molecular-weight hexamers and subsequently assemble into high-molecular-weight oligomers exceeding 300 kDa, the latter being considered the most biologically active isoforms. Inter-assay Precision (precision between assays): Three samples of known concentration were tested six times on different plates to assess inter-assay precision. Spike Recovery The spike recovery was evaluated by spiking three levels of human adiponectin into healthy human serum samples. The unspiked serum was used as a blank in this experiment. The recovery ranged from 81% to 126%, with an overall mean recovery of 93%.
