Juice™ minibinder cell-free protein expression system is the only kit on the market specifically optimized for minibinder expression. The mix can produce ~1 mg of purified minibinder constructs per milliliter of reaction in under 16 hours starting from optimized linear templates. It has been designed for T7-based protein expression and can generate proteins from standard inducible expression plasmids (e.g. pET15 and pET28) in the presence of 0.5-1mM IPTG. An important advantage of the Juice system is that it does not contain polyethylene glycol (PEG). PEG, which is often present in other commercially available cell-free systems, is a hydrophobic agent that can result in ion suppression at the ion source of a mass spectrometer and appears as repeating elements of 44 Da in the mass spectrum (Scheerlinck et al, 2015).
