Juice™ nanobody cell-free protein expression system is the only kit on the market specifically optimized for nanobody, VH or VHH expression. The mix can produce over 1.4 mg of purified nanobody constructs per milliliter of reaction in under 16 hours starting from optimized linear templates. It has been designed for T7-based protein expression and can generate proteins from standard inducible expression plasmids (e.g. pET15 and pET28) in the presence of 0.5-1mM IPTG. An important advantage of the Juice system is that it does not contain polyethylene glycol (PEG). PEG, which is often present in other commercially available cell-free systems, is a hydrophobic agent that can result in ion suppression at the ion source of a mass spectrometer and appears as repeating elements of 44 Da in the mass spectrum (Scheerlinck et al, 2015).
