This end-to-end automated workflow takes Pichia pastoris glycerol stocks through three sequential preculture and production fermentation stages, then quantifies secreted protein ex pression by size and purity via microfluidic capillary electrophoresis on the Revvity LabChip. Automation spans 384-well high-throughput transformation using the Agilent Bravo 96 and Inheco ATC Block, multi-stage fed-batch-style growth in the Cytomat incubator (30°C, 1000 rpm, 80% humidity), supernatant harvest by high-speed centrifugation, and LabChip-based protein quantification.
