Hieff NGSTM dsDNA Methyl Library Prep Kit for IlluminaTM is a double-stranded DNA methylation library kit. This kit can construct a methylation library for Illumina platform sequencing by connecting methylated adapters to DNA that has been treated with bisulfite or enzymatic conversion, and then amplifying it in one step. All components of the kit have passed quality control and functional verification to ensure stable output of the library, and support a variety of DNA samples, including gDNA, cfDNA, and FFPE. This kit is compatible with 10 ng-1000 ng DNA and is compatible with mainstream conversion schemes on the market. Features Easy operation: The double-stranded methylation library workflow is simpler than single-stranded library preparation, requiring one less adapter ligation and purification step. Simplified analysis: Double-stranded methylation library data analysis does not require read trimming, resulting in more usable data. High hybrid capture mapping: Hybrid capture achieves high mapping rates without the need for special blocking reagents. Using HieffTM Mag DNA Methylation Bisulfite Kit(Cat#12223), gDNA, cfDNA, and FFPE samples with inputs of 10 ng, 100 ng, and 1 μg were prepared. Library yields outperformed those from Supplier A* across all input amounts, demonstrating consistent performance across different sample types. 3. Broad compatibility: Supports both BS and EM-seq workflows. Figure 3. Robust performance across methylation conversion methods. Using Cat#12214, dsDNA libraries prepared from 10–200 ng DNA input demonstrate conversion efficiency comparable to ssDNA library prep(Cat#12221), while supporting both bisulfite (BS) and enzymatic methylation (EM) workflows. 4. Excellent Sequencing Data Performance Figure 4. Yeasen Kit (Cat# 122214) delivers superior library yield and data quality after bisulfite (BS) conversion. Across various DNA templates, libraries prepared with Yeasen 122214 showed higher yields and better data quality compared to those generated using alternative methods.
