This end-to-end automated workflow combines cell-free protein synthesis (CFPS) with Strep-tag II affinity purification and LabChip-based protein quantification. Linear DNA templates are expressed in CFPS reactions in 96-well deep-well plates, with reagents dispensed via BioTek MultiFlo. Expressed proteins bearing a Strep-tag II are purified using magnetic bead-based affinity purification on the Agilent Bravo 96. Purified eluates are then diluted, transferred, and analyzed on the Revvity LabChip system for quantitative assessment of expression, titer, and purity. The protocol supports up to 88 constructs per run (with 1 column reserved for controls) and delivers per-sample expression confirmation at expected molecular weight, baseline titer (mg/L), and initial purity with virtual gel images.
