IL-27 (Interleukin 27) is a cytokine belonging to the IL-12 family, composed of an EBI3 and a p28 subunit forming a heterodimeric cytokine. IL-27 is produced by activated dendritic cells and macrophages in response to Toll-like receptor ligands and pro-inflammatory cytokines. IL-27 promotes the differentiation of CD4+ T cells into the Th1 lineage by inducing the expression of the transcription factor T-bet and upregulating IL-12Rβ2, which in turn inhibits the differentiation and proliferation of Th2 and Th17 cells. IL-27 is a recently identified member of the pro-inflammatory and immunoregulatory IL-6/IL-12 cytokine family. IL-27 functions by binding to its receptors, WSX-1 and gp130, leading to the activation of the Jak/STAT signaling cascade, indicating that IL-27 is involved in the regulation of immune processes.
